The effect of FK506 on the pemphigus vulgaris (PV) sera-induced phosphorylation of both p38 mitogen-activated protein kinases (MAPK) and heat shock protein (HSP) 27. HaCaT cells were pretreated with 100 nM FK506 or 1 µM SB203580 (p38 MAPK inhibitor) for 2 h, followed by exposure to 5% PV sera or 1 µg/ml AK23 for 1 h. FK506 significantly blocked the phosphorylation of HSP27, but not p38 MAPK. Levels of total p38 MAPK are shown as loading controls (n = 3). (B and C) Data are represented by mean ± SE.
